Deze tweede druk komt grotendeels overeen met de eerste. Maar om het boek aan te passen aan de huidige stand van de wetenschap en de technologie zijn er toch diverse belangrijke wijzigingen aangebracht. Zo is hoofdstuk 21 helemaal aangepast aan de nieuwste richtlijnen voor immuuntypering van maligne hematologische ziekten. De hoofdstukken 23 en 24 hebben veel vernieuwingen ondergaan die zijn ingegeven door de zich snel uitbreidende inzichten op het gebied van (patho)fysiologie van trombocyten, stollingsfactoren en hun remmers. Omdat het endotheel van de vaatwand functioneel zo nauw verbonden is met trombocyten en stollingsfactoren wordt het niet meer in een apart hoofdstuk behandeld, maar is de tekst opgenomen in de hoofdstukken 23 en 24. De introductie van nieuwe geneesmiddelen tegen trombose maakte enkele wijzigingen in hoofdstuk 28 nodig. Verscheidene hoofdstukken in deel 5 zijn volledig aangepast aan de in 2004 verschenen CBO Richtlijn Bloedtransfusie. Tenslotte zijn de appendices geheel vernieuwd.
